EPR STUDIES ON NITRITE REDUCTION TO NITRIC OXIDE IN Pseudomonas BACTERIA

چکیده
Introduction The reduction of nitrite to nitric oxide (NO) by whole cells of denitrifying bacteria Pseudomonas aeruginosa, P. jluorescens, P. aerufaciens and Acaligenes fecalis was studied by EPR in conjunction with spin trapping technique. Among these bacteria, P. aeruginosa which is ubiquitous in nature and generally innocuous in most environments, is of special clinical interests, since it can cause severe infections in immunosuppressed patients suffering fiorn burn, respiratory disease or cystic fibrosis. We have confirmed that these various Pseudomonas bacteria, under anaerobic condition, are able to reduce nitrite to NO by the enzyme nitrite reductase. It is well known that NO, a very reactive molecule, is capable of giving rise to toxic radical species, thus overproduction of NO contributes to numerous pathological conditions.

The requirement of RpoN (sigma factor sigma54) in denitrification by Pseudomonas stutzeri is indirect and restricted to the reduction of nitrite and nitric oxide.
The rpoN region of Pseudomonas stutzeri was cloned, and an rpoN null mutant was constructed. RpoN was not essential for denitrification in this bacterium but affected the expression levels and enzymatic activities of cytochrome cd1 nitrite reductase and nitric oxide reductase, whereas those of respiratory nitrate reductase and nitrous oxide reductase were comparable to wild-type levels. Since t...

Studies on the process of nitrite reduction by cell-free extracts of denitrifying bacteria could materially contribute to our knowledge of its mechanism. Such investigations have so far been limited to those of Yamagata (1) with Bacillus pyocyaneus (Pseudomonas aeruginosa) and of Robinson et al. (‘2) with Micrococcus haZodenitri$cans. Neither of these authors has examined the products of the re...

متن کاملCatalase (KatA) Plays a Role in Protection against Anaerobic Nitric Oxide in Pseudomonas aeruginosa
Pseudomonas aeruginosa (PA) is a common bacterial pathogen, responsible for a high incidence of nosocomial and respiratory infections. KatA is the major catalase of PA that detoxifies hydrogen peroxide (H2O2), a reactive oxygen intermediate generated during aerobic respiration. Paradoxically, PA displays elevated KatA activity under anaerobic growth conditions where the substrate of KatA, H2O2,...
متن کامل15N tracer studies on the reduction of nitrite by the purified dissimilatory nitrite reductase of Pseudomonas aeruginosa. Evidence for direct production of N2O without free NO as an intermediate.
Nitrite reductase (cytochrome c,d1) was purified from Pseudomonas aeruginosa. In the presence of the reducing system, ascorbate-N,N,N',N'-tetramethylphenyl-enediamine, which alone had no ability to reduce nitrite or NO at pH 7.5, the enzyme catalyzed the reduction of nitrite to NO and N2O as major and minor products, respectively, as determined by gas chromatography-mass spectrometry. The rate ...
